Tulisan ini bertujuan mengkaji makna persaudaraan spiritual dalam komunitas umat Kristiani berdasarkan teks Kisah Para Rasul 2:1 serta relevansinya bagi kehidupan rohani mahasiswi Komunitas Asrama Skolastika dan Benediktus Stipar Ende. Metode yang digunakan adalah studi kepustakaan (library research) dengan pendekatan hermeneutik biblis dan deskriptif-kualitatif. Teks Kisah Para Rasul 2:1 menggambarkan peristiwa Pentakosta sebagai momen lahirnya komunitas iman yang diikat oleh Roh Kudus dalam kesatuan, kesehatian, dan persekutuan doa. Makna persaudaraan spiritual yang terkandung di dalamnya mencakup dimensi vertikal (hubungan dengan Allah melalui Roh Kudus) dan dimensi horizontal (hubungan antaranggota komunitas dalam kasih, pengorbanan, dan pelayanan bersama). Relevansi temuan ini bagi mahasiswi asrama ditunjukkan melalui tiga aspek utama: (1) penguatan identitas komunitas rohani, (2) pembentukan karakter Kristiani dalam kehidupan bersama, dan (3) pengembangan spiritualitas komunal yang berakar pada tradisi apostolik. Hasil kajian menegaskan bahwa persaudaraan spiritual bukan sekadar ikatan sosial, melainkan panggilan ilahi yang menuntut keterbukaan terhadap karya Roh Kudus, kerendahan hati, dan komitmen terhadap kehidupan doa serta pelayanan bersama dalam komunitas asrama.
